(Skilled detect treasure is just blessed object detection: it DOES NOT reveal the appearance of monster inventories.)
(Ruthlessly deleted the material duplicated from the potion of object detection page and linked to it: duplication is bad. Fixed the strategy section: detect treasure is good.)

Treasures detected are always real. If NetHack doesn't show the gold/gem symbol when you get to the marked spot, it means that either it was in the inventory of a monster (that could have moved), or the items are buried in the floor. In the latter case, just dig a pit (apply a pickaxe down >) and you'll find the stuff.

Although mostly useless, the detect treasure spell allows you to remotely detect vaults, which appear as four gold coins (provided they haven't been looted by you, or eaten by a rock mole):

The spell is also sometimes used in the Castle, as an easy way to detect where the chest containing the wand of wishing is. It has some use in Gehennom, where, especially when combined with magic mapping and/or teleport, it allows you to traverse through the levels having picked everything up without fully navigating the maze.

Allows you to learn the detect treasure spell. This spell has the same effect as a potion of object detection.[1] It is the special spell for rogues.

Casting effects

Case Effect
Unskilled or Basic Same effect as quaffing an uncursed potion of object detection. See that page.
Skilled or Expert Same effect as quaffing a blessed potion of object detection. See that page.

Strategy

The number of potions you can use is limited by the number you can find and carry. The spell however is weightless and unlimited, this makes it much more useful. Since the spell is non-combat it is usable even with a fairly high failure rate. Even better, it shares it's spell school (divination) with magic mapping and identify, two other excellent spells.

If you can, you should use this spell.

 1. spell.c, line 910 a fake potion is loaded up and used, allowing the peffects (potion effects) code to be reused for spell effects. Thus the spell and the potion are implemented by the same code.
