In NetHack, the letter S serves the following purposes:

Historical uses

In Hack 1.21, based on Jay Fenlason's Hack, stalactoids use the s glyph, while snowmen use the S glyph. In Hack for PDP-11, the l is given to the giant scorpion, while the S glyph is given to the snake - the snake retains its glyph for its appearance in Hack 1.0, while the giant scorpion is replaced with the regular scorpion. Both monsters continue to use those glyphs up to NetHack 2.3e.

In NetHack 3.0.0, the arachnid or centipede monster class is introduced and uses the s glyph, with the scorpion made part of the monster class, while the snake monster class is introduced and given the S glyph.
